چکیده

Suppression of PentylenetetrazolInduced Seizures and c-fos Expression in Mouse Brain by L-Carnitine: Yoshihisa IRYO, et al. Department of Environmental Toxicology, Institute of Industrial Ecological Sciences, University of Occupational and Environmental Health—Seizure development in ddY mice was recorded on a videotape for 20 min after injection of pentylenetetrazol (PTZ) (50 mg/kg) and analyzed in detail. In addition, expression of the c-fos gene in the brain was semi-quantified by reverse transcription-polymerase chain reaction analysis. When saline, L-carnitine (10 mmol/kg), or Dcarnitine (10 mmol/kg) was administered 30 min prior to PTZ, the seizures were suppressed in mice given Lcarnitine; the seizure scores were significantly lower in mice treated with L-carnitine at several time points, and the overall seizure scores were 31.43 ± 2.49, 10.57 ± 2.86 and 24.71 ± 3.05 (expressed as the mean ± S.E.M., n=7), in saline, L-carnitine and D-carnitine groups, respectively. The latency to the first clonictonic seizure was also prolonged in mice treated with L-carnitine. The level of c-fos mRNA in the brain was lower in the animals treated with L-carnitine than in those treated with saline or D-carnitine. Thus, Lcarnitine shows not only anticonvulsive effects in one of the most widely used animal models of chemically induced seizures but also the potential to suppress the seizure-associated expression of an immediate early gene in the brain. Anticonvulsive and antioxidant effects of pioglitazone on pentylenetetrazol-induced seizures in mice

Introduction: Epilepsy is a neurological disorder caused by uncontrollable discharge of neurons in the brain. After a seizure oxidative stress play an important role in the oxidative neuronal damage. In this study, we evaluated the anticonvulsant and antioxidant effects of pioglitazone, a PPAR-γ agonist that is used in type2 diabetes, on pentylenetetrazol-induced seizure in mice. Methods: In t...

Modulation of c-Fos and BDNF Protein Expression in Pentylenetetrazole-Kindled Mice following the Treatment with Novel Antiepileptic Compound HHL-6

Brain-derived neurotrophic factor (BDNF) and c-Fos are shown to promote epileptogenesis and are taken as a marker of neuronal activity. The present study investigated the expression of BDNF and c-Fos in mice brain with pentylenetetrazol- (PTZ-) induced generalized seizure and evaluated the effect of novel tryptamine derivative HHL-6 on the expression of these two markers. The subconvulsive dose...

Study the Efect of Retinoeic Acid and Mitogenes on Lens and Optic Vesicle of Mouse Embryo(Nmri) and Fos Protein Expression In Culture Medium

Purpose: This work studies possible role of FOS proteins, retinoic acid (RA) and mitogens during optic vesicle development in vitro. Materials and Methods: Fos was detected as a product of 55-62 kDa in optic vesicles via electrophoretic procedures. Incubation (24hr) of optic vesicles with RA 25nM and mitogens(serum10%,insulin2.5%) on the day 9.5 of gestation revealed that Fos was induced by mi...

Intrathecal Amylin and Salmon Calcitonin Affect Formalin Induced c-Fos Expression in the Spinal Cord of Rats

Background: Amylin and Salmon Calcitonin belong to the calcitonin family of peptides and have high affinity binding sites in the rat spinal cord. The aim of this study was to characterize receptors for Amylin and Salmon Calcitonin functionally in the spinal cord of rats. We assessed the expression of c-Fos in response to intraplantar formalin in the lumbar regions of the spinal cord in consciou...

Anticonvulsant Activity of Viola tricolor against Seizures Induced by Pentylenetetrazol and Maximal Electroshock in Mice

Background: Recently, there has been much more interest in the use of medicinal plants in search of novel therapies for human neurodegenerative diseases such as epilepsy. In the present study, we investigated the anticonvulsant effects of Viola tricolor (V. tricolor) on seizure models induced by pentylenetetrazol (PTZ) and maximal electroshock stimulation (MES). Methods: Totally, 260 mice were ...
